ՄիացյալՆահանգների, Բրազիլիայի, Իրանի և Իսրայելի գիտնականները պարզել են, որ նիհարելու համար նախատեսված ներարկային դեղամիջոցները, մասնավորապես՝ «Օզեմպիկ»-ը, մեծացնում են դեպրեսիայի և ինքնասպանության մտքերի ռիսկը դոֆամինի որոշակի տեսակի ակտիվություն ունեցող մարդկանց շրջանում: Հետազոտության արդյունքները հրապարակվել են Current Neuropharmacology (CurNeuro) ամսագրում:
«Ozempic»-ը և նրա անալոգները նմանակում են գլյուկագոնման պեպտիդ-1 (GLP-1) հորմոնի աշխատանքը: Ամեն անգամ կերակուր ընդունելուց հետո ստամոքսում GLP-1-ն արտազատվում է: Այս հորմոնն օգնում է ավելի շատ ինսուլին արտադրել՝ ակդկեպ իջեցնելով արյան մեջ գլյուկոզի մակարդակը և դանդաղեցնելով ստամոքսի դատարկման արագությունը:
Թիմն օգտագործել է առաջադեմ դեղաբանա-գենետիկական տեխնիկա՝ գենետիկական առանձնահատկությունները բացահայտելու համար, որոնք կարող են որոշ մարդկանց ավելի խոցելի դարձնել այս դեղերի հոգեբուժական կողմնակի ազդեցությունների նկատմամբ: Հետազոտությունը ցույց է տվել, որ GLP-1 ընկալիչների վրա գործող դեղամիջոցները կարող են տարբեր կերպ ազդել մարդկանց վրա՝ կախված նրանց դոֆամինային ակտիվությունից: Այսպես, դոֆամինային համակարգի (հիպոդոպամիներգիա) գործառույթի նվազեցված բուժառուների շրջանում մեծանում է դեպրեսիվ ախտանիշների առաջացման վտանգը:
Հեղինակները միաժամանակ կապ են հայտնաբերել այս դեղամիջոցների ընդունման և տրամադրությունը և մոտիվացիան կարգավորող հիմնական գեների՝ DRD3, BDNF և CREB1 փոփոխությունների միջև: Այդ գեների խախտումը կարող է նպաստել դեպրեսիայի և ինքնասպանության վարքագծի զարգացմանը:
