Վաշինգտոնի Ֆրալինի կենսաբժշկական հետազոտությունների ինստիտուտի քաղցկեղի ուսումնասիրման կենտրոնի հետազոտողները պոտենցիալ թիրախ են բացահայտել փորձարարական դեղամիջոցների համար, որոնք արգելափակում են PRMT5-ը՝ բնական ֆերմենտ, որից մեծապես կախված է որոշ ուռուցքների կենսունակությունը։

Cancer Research ամսագրում հրապարակված հետազոտության մեջ Ֆրալինի կենսաբժշկական հետազոտությունների ինստիտուտի դոցենտ Քեթլին Մալվանին ներկայացրել է հետազոտության արդյունքները, որոնք կարող են օգնել ստեղծել թոքերի, գլխուղեղի և ենթաստամոքսային գեղձի քաղցկեղի որոշ տեսակների բուժման նոր մեթոդներ։

Թոքերի քաղցկեղն ամբողջ աշխարհում քաղցկեղից մահացության հիմնական պատճառն է։ Ենթաստամոքսային գեղձի քաղցկեղով հիվանդների հինգամյա կենսունակությունը 15 տոկոսից պակաս է, իսկ գլիոբլաստոմայի դեպքում՝ էլ ավելի ցածր։

Բուժումը հաճախ անհաջող է լինում։ Ստացված տվյալները վկայում են, որ PRMT5-ի արգելակիչը կարող է դառնալ դժվար բուժվող որոշ քաղցկեղների բուժման նոր արդյունավետ մեթոդ։

Այս ուռուցքներից շատերն ունեն ընդհանուր գենետիկական առանձնահատկություն. դրանցում բացակայում են CDKN2A-ն և MTAP-ը՝ երկու գեներ, որոնք ճնշում են ուռուցքների աճը և օգնում կարգավորել բջիջների աճը։ Առանց դրանց քաղցկեղային գոյացությունները կախվածության մեջ են ընկնում PRMT5-ից և պոտենցիալ խոցելի դառնում այս ֆերմենտն արգելափակող դեղամիջոցների նկատմամբ։ Մալվանին և գործընկերները վերլուծել են հազարավոր ուռուցքաբանական հիվանդների գենետիկական տվյալները, որոնք հասանելի են cBioPortal պորտալում։

Նրանք կիրառել են CRISPR խմբագրման գործիքներ՝ ուսումնասիրելու կենսաբանական ուղիները տարբեր նմուշներում՝ պարզելու, թե որ գեներն են քաղցկեղի բջիջներն ավելի խոցելի դարձնում PRMT5-ի արգելակիչների նկատմամբ, և որ համակցությունները կարող են բարելավել արձագանքն ու երկարաժամկետ արդյունքները։

Ըստ Մալվանիի գնահատականների, քաղցկեղով հիվանդների մոտ 5 տոկոսը՝ տարեկան 80 000-ից 100 000 մարդ ԱՄՆ-ում, կարող է օգուտ քաղել առաջարկվող բուժման մեթոդներից։

«Հայտնաբերել ենք նաև մի շարք գեներ, որոնք քաղցկեղի դեպքում փոխազդում են PRMT5 ազդանշանի հետ, ինչի մասին նախկինում հայտնի չէր», - ասել է Մալվանին։

Ի լրումն թոքերի, գլխուղեղի և ենթաստամոքսային գեղձի քաղցկեղի, սա հեռանկարային բուժում է մելանոմայի և մեզոթելիոմայի որոշ տեսակների համար։ Լաբորատորիայի աշխատակիցները հաջողության են հասել պոտենցիալ բուժման մեթոդները փորձարկելուց հետո՝ ինչպես կենդանիների մոդելների, այնպես էլ հիվանդների հյուսվածքներից ստացված բջջային կուլտուրաների վրա։
