Գիտնականները ուղեղում քաղցի և քաշի վերահսկման նոր մեխանիզմ են բացահայտել

Լայպցիգի համալսարանի և Charité – Universitätsmedizin Berlin-ի գիտնականները հայտնաբերել են հիմնական մեխանիզմը, որը կարգավորում է ախորժակը և քաշի վերահսկողությունը, ինչն օգնում է ուղեղին կառավարել քաղցի զգացումը։ Collaborative Research Centre (CRC) 1423 – Structural Dynamics of GPCR Activation and Signaling-ի շրջանակներում անցկացված հետազոտության մեջ գիտնականները պարզել են, թե ինչպես է MRAP2 (melanocortin 2 receptor accessory protein 2) սպիտակուցն ազդում MC4R (melanocortin-4 receptor) ուղեղային ընկալիչի աշխատանքի վրա, որը կենտրոնական դեր է խաղում ախորժակի և էներգետիկ հաշվեկշռի վերահսկման գործում։
Արդյունքները հրապարակվել են Nature Communications ամսագրում։
MC4R ընկալիչն ակտիվանում է MSH պեպտիդային հորմոնով և մասնակցում է քաղցի զգացումի կարգավորմանը։ Այս ընկալիչի մուտացիաները համարվում են ծանր ճարպակալման ամենատարածված գենետիկ պատճառներից մեկը։ Նախկինում հետազոտողները վերծանել էին ակտիվ ընկալիչի եռաչափ կառուցվածքը՝ կոմպլեքսում լիգանդների և սետմելանոտիդ դեղամիջոցի հետ, որը նվազեցնում է քաղցի զգացումը։ Սա հնարավորություն տվեց ավելի լավ հասկանալ MRAP2-ի և բջիջների մակերեսին ընկալիչի տեղափոխման գործում նրա դերի մասին նոր ֆունկցիոնալ տվյալները, որտեղ այն կարող է ավելի արդյունավետ ճնշել ախորժակը։
Օգտագործելով ժամանակակից ֆլուորեսցենտային մանրադիտակություն և մեկբջիջային պատկերում՝ թիմը ցույց է տվել, որ MRAP2-ը զգալիորեն փոխում է MC4R-ի տեղակայումը և վարքը բջիջների ներսում։ Ֆլուորեսցենտային կենսազգացիչները և կոնֆոկալ մանրադիտակությունը ցույց են տվել, որ MRAP2-ը անհրաժեշտ է MC4R-ի՝ բջջային մակերես տեղափոխելու համար՝ ապահովելով ախորժակը ճնշող ազդանշանների ավելի արդյունավետ փոխանցում։
Ընկալիչի կարգավորման նոր մակարդակի հայտնաբերումը մատնանշում է հեռանկարային թերապևտիկ ռազմավարություններ, որոնք նմանակում կամ մոդուլացնում են MRAP2-ը, ինչը կարող է օգնել պայքարել ճարպակալման և նյութափոխանակության խանգարումների դեմ։ Գերմանիայի, Կանադայի և Միացյալ Թագավորության հետազոտողների միջդիսցիպլինար և միջազգային թիմը համատեղել է գիտելիքները կենդանի բջիջների մանրադիտակության, մոլեկուլային դեղաբանության և կառուցվածքային կենսաբանության ոլորտներում՝ ցուցադրելով ընկալիչների կարգավորման ուսումնասիրության համալիր մոտեցման արդյունավետությունը։
