Վիսկոնսին-Մեդիսոնի համալսարանի գիտնականները հայտնաբերել են, որ մեր քրոմոսոմների կայունության համար պատասխանատու սպիտակուցի աշխատանքի խանգարումները կարող են լինել լուրջ և երբեմն մահացու հիվանդությունների պատճառ։

Հետազոտության արդյունքները հրապարակվել են Science ամսագրում և կարող են օգնել բժիշկներին ու հիվանդներին բացահայտել սպիտակուցների նոր մուտացիաներ, որոնք կապված են քաղցկեղի որոշակի տեսակների և ոսկրածուծի հիվանդությունների հետ։

Քրոմոսոմները ԴՆԹ-ի և սպիտակուցների «փաթեթավորված» կոմպլեքսներ են, որտեղ պահվում է մեր ամբողջ գենետիկ տեղեկատվությունը։ Դրանց ծայրերը պաշտպանված են թելոմերներով՝ ԴՆԹ-ի և սպիտակուցների կրկնվող հաջորդականություններից բաղկացած յուրօրինակ գլխարկներով։ Թելոմերները բնականաբար կարճանում են տարիքի հետ, սակայն դրանց ձևավորման կամ պահպանման խափանումները ԴՆԹ-ն ավելի պակաս կայուն են դարձնում՝ հանգեցնելով վաղաժամ ծերացման և հիվանդությունների։

Կենսաքիմիայի պրոֆեսոր Ցի Ջի Լիմի գլխավորությամբ հետազոտողները քիմիայի ամբիոնի գիտնականների հետ միասին ուսումնասիրել են այն սպիտակուցները, որոնք փոխազդում են թելոմերազի՝ թելոմերների երկարությունը պահպանող ֆերմենտի հետ։ Այդ սպիտակուցների աշխատանքի խափանումը կարող է առաջացնել կարճացած թելոմերների հետ կապված հիվանդություններ։

AlphaFold-ի՝ սպիտակուցների եռաչափ կառուցվածքը և դրանց փոխազդեցությունները կանխատեսելու մեքենայական ուսուցման համակարգի օգնությամբ, թիմը բացահայտել է replication protein A (RPA) սպիտակուցը՝ որպես թելոմերների պահպանման հիմնական տարր։ RPA-ն վաղուց հայտնի է որպես ԴՆԹ-ի կրկնօրինակման և վերականգնման մասնակից, սակայն մարդկանց մոտ երկար և առողջ թելոմերների պահպանման գործում նրա դերը նախկինում չի հաստատվել։ Փորձարարական ստուգումները ցույց են տվել, որ RPA-ն իսկապես անհրաժեշտ է թելոմերազը խթանելու և թելոմերները պահպանելու համար։

Այս հայտնագործությունն անմիջական նշանակություն ունի կարճացած թելոմերներով պայմանավորված վտանգավոր հիվանդություններով տառապող հիվանդների համար, ներառյալ ապլաստիկ անեմիան, միելոդիսպլաստիկ համախտանիշը և սուր միելոիդ լեյկոզը։ Ըստ Լիմի, նախկինում թելոմերների խանգարումներ ունեցող մի շարք հիվանդների համար հայտնի տվյալների հիման վրա բացատրություն չկար, իսկ այժմ նրանք հասկանում են, որ որոշ մուտացիաների պատճառը RPA-ի՝ թելոմերազը խթանելու անկարողությունն է։

Գիտնականներն արդեն հարցումներ են ստացել ամբողջ աշխարհի բժիշկներից և հետազոտողներից՝ արդյոք իրենց հիվանդների հիվանդությունները կապված չեն գենետիկ մուտացիաների հետ, որոնք արգելափակում են RPA-ի ֆունկցիան։
