Հայտնաբերվել են չարորակ ուղեղի ուռուցքներ առաջացնող բջիջներ

Յոնսեյ համալսարանի գիտնականները հայտնաբերել են, որ ուղեղի ուռուցքները կարող են առաջանալ նորմալ բջիջներից, որոնք վաղ փուլում ձեռք են բերում վտանգավոր գենետիկ մուտացիա և երկար ժամանակ մնում են չհայտնաբերված: Միայն տարիներ անց են այս փոփոխված բջիջները վերածվում տեսանելի ուռուցքների: Ուսումնասիրության արդյունքները հրապարակվել են Nature ամսագրում :
Գիտնականները կենտրոնացել են IDH գենի մուտացիայով չարորակ գլիոմայի վրա, որը 50 տարեկանից փոքր մարդկանց մոտ ուղեղի քաղցկեղի ամենատարածված և դժվար բուժվող տեսակներից մեկն է: Հիվանդությունը սկսում է զարգանալ տեսանելի ուռուցքային զանգվածի ի հայտ գալուց շատ ավելի շուտ, ինչը նշանակում է, որ բուժման ներկայիս մոտեցումները կարող են բաց թողնել դրա դիսեկցիայի կարևոր փուլը:
Հետազոտողները վերլուծել են ուռուցքային հյուսվածքի նմուշներ և ակնհայտորեն առողջ ուղեղային կեղև, որոնք վերցվել են լայնածավալ նյարդավիրաբուժական միջամտությունների ընթացքում: Նրանք հայտնաբերել են IDH մուտացիայով բջիջներ նորմալ համարվող հատվածներում: Այս բջիջներն արդեն կրում էին «առաջնային» արատը, բայց դեռ չէին ձևավորել լիարժեք ուռուցք:
Հետագա փորձարկումները ցույց տվեցին, որ գլիալ նախորդ բջիջները կրում են վաղ մուտացիան: Այս բջիջները առկա են նորմալ ուղեղում և, որոշակի գենետիկական արատների դեպքում, կարող են դառնալ չարորակ պրոցեսների սկզբնակետ: Մկների վրա կատարված փորձարկումները հաստատեցին այս արդյունքները. նույն IDH մուտացիան այս բջիջների մեջ ներմուծելը խթանեց ուռուցքի զարգացումը:
«IDH-մուտացված գլիոմաները չեն առաջանում հանկարծակի, այլ զարգանում են դանդաղ՝ երկար ժամանակահատվածում, տարածվելով ամբողջ ուղեղային կեղևով մեկ։ Սա կարող է բացատրել տեսանելի ուռուցքային հյուսվածքի վիրաբուժական հեռացումից հետո հաճախակի կրկնությունները», - նշել են հեղինակները։
Գիտնականները շեշտել են, որ ուղեղի քաղցկեղի տարբեր տեսակներ ունեն զարգացման տարբեր մեխանիզմներ: Նրանք ասում են, որ ուռուցքից առաջացող բջիջների վրա կենտրոնանալը, այլ ոչ թե միայն ուռուցքի վրա, կարող է հիմք հանդիսանալ վաղ ախտորոշման և բուժման ռազմավարությունների համար:
